Method :
Bactec® assay system (tested duration for 1day - 25 days)

Mycobacterial plate counts from Bactec® vials :
Bacterial plate counts were done on various Bactec vials to determine the viability of mycobacteria exposed to WAP-8294A2. 1Χ104-5 bacteria were injected into each Bactec vial at the start to the run. Then the antibiotic was injected into the vials. Bactec readings were taken daily. At certain time points during the run vials were removed to determine the number of colony forming units (CFU) in the vial. Broth cultures were transferred from the Bactec vials to tubes containing glass beads. The beads when the tubes are vortexed for a few seconds help to break up the clumps of mycobacteria-allowing for a more accurate CFU determination. The cultures were serially diluted in 7H9 containing Tween 80 to prevent clumping, plated on 7H11, and incubated. Two vials from a Bactec run screening M. avium were counted.

Results:
1. No cross resistance against anti-tuberculosis agent strains.
2. WAP-8294A2 may have bactericidal activity.
3. Growth inhibition of WAP-8294A2 was indicated for 2 - 8 mcg/ml of concentration.
